Contribution of the solid-state loading method of molecular materials within mesoporous silica matrices: from pharmaceutical application to the exploration of physical state transformations of confined matter
Abstract :
Active pharmaceutical ingredients are most often synthesized in crystalline form. This state is stable but has a lower bioavailability compared to an amorphous state. Confining an active ingredient within mesoporous silica matrices is one strategy to stabilize its amorphous state and control its release. The development of this strategy has been the subject of several studies aiming to address issues related to drug formulation. The methods used for loading primarily rely on using sometimes toxic solvents or melting the active pharmaceutical ingredient. Both of these methods are incompatible with therapeutic applications. Milling-assisted loading is a recent method developed to overcome the drawbacks of classic loading methods. This thesis work focused on optimizing this loading method and also extending its scope to biomolecular systems.
